To do this properly, you need to multisample whatever it is you want to play up and down the keyboard. Couple of ways to solve this. Your bottom end ( <150) should probably stay mono. Layer a mono subbier kick and a stereo mid/click one. Reverb (as already mentioned) is another way. Having read all of that, yes, there can be a difference. Comp samplers reproduce what you put into them. The EMU stuff was always good kit. The E4X are kind of the pinnacle of hw samplers.
